Cabbage (Brassica oleraceae var. Capitata L.) is a cross-pollinated agricultural crop propagated by seeds. In this study were used two varieties of cabbage 'Ogulinski' and 'Varaždinski' in order to examine the possibility of using in vitro culture in the preservation of old varieties that are inadequately maintained (resulting in loss of varietal characteristics typical of each variety) or are on the verge of extinction. In the study of regeneration of 'Ogulinski' and 'Varaždinski' cabbage in in vitro culture, seed germination was performed on MS (Murashige and Skoog, 1962) medium. Higher germination of 'Ogulinski' cabbage seeds was recorded compared to 'Varaždinski' cabbage. Regeneration was performed from the hypocotyl (basal and apical) and petioles on two nutrient media (MS with thidiazuron or 6-benzylaminopurine). 'Ogulinski' cabbage proved to be a variety with more significant morphogenicity than 'Varaždinski' cabbage, as well as explants obtained from the basal hypocotyl compared to the apical hypocotyl . The percentage of morphogenic explants obtained from the petioles of 'Varaždinski' (11.2%) and 'Ogulinski' (16.2%) cabbages did not differ significantly. MS media with auxins NAA, IBA, IAA and hormone-free were used in the rooting phase. A significant difference in rooting percentage was found between nutrient media with different hormones and between varieties. The rooting results indicate a significantly higher rooting of 'Ogulinski' cabbage compared to 'Varaždinski' cabbage. Acclimatization of plants rooted on medium with NAA was not successful, and there were no surviving plants in any variety, while plants rooted on medium suplemented with IBA all survived. Cabbage micropropagation could be used for the purpose of preserving varieties of cabbage whose seeds are insufficient and very poorly germinated, which may lead to the final loss of the variety.
