The Caper bush (Capparis spinosa L.) is wild decidous perrenial plant, in Croatia growing on mostly karst areas of Dalmatia. The caper is rich in nutrients and year after year increasing its economic value. Seed of caper is highly dormant and haterozygous, so the most important increase in caper plants for market production is micropropagation, resulting in a large number of genetically uniform plants. Previous studies have shown that the caper plant can be very successfully multiplied in vitro by axillary branching. The purpose of this master’s thesis was to investigate how the concentration of macro- and micro- salts affects in vitro rooting of capers, to find out optimal time length of the rooting, to investigate the different auxins and their concentrations on the rooting of the micropropagated shoots, also to consider influence of the activated charcoal, the carry-over effect and the rate of acclimatization. Research results show that higher concentrations of macro- and micro- salts give a higher percentage of in vitro rooted capers. The 90-day rooting duration gives a higher number of rooted capers (85%), and the highest percentage of rooted caper plants is obtained on a medium of IAA (indol-3-acetic acid) 2 mg/L. The largest root length appears on the medium without growth hormone and in the presence of activated charcoal. The highest rate of caper acclimatization (65,78%) was obtained on a medium containing IBA (indol-3-butiric acid) 2 mg /L.
