| Sažetak (engleski) | The raw experimental data which was analysed and published in the manuscript '' Tyrosyl-DNA phosphodiesterase 2 (Tdp2) repairs DNA-protein crosslinks and protects against double strand breaks in vivo'' is uploaded in this repository. The raw data include enzymatic actvity data in .xlsx, .pzfx, and .tif. RTPCR results in .xlsx and .pzfx; images of zebrafish embryos in .jpg and .tif; visualization and quantification of DNA-protein crosslinks and γH2AX western blots in .xlsx, .pzfx and .tif. |
| Metodologija (engleski) | DPC isolation was performed using modified RADAR (rapid approach to DNA adduct recovery) assay. DPCs were separated using SDS-PAGE electrophoresis and visualized using silver staining. Specific DPCs were visualized using dot blot analysis and detected with specific antibodies. yH2AX levels were visaulized using Western blot. Quantification of silver-stained gels, Dot blots and Western blots was conducted using the ImageJ software. Zebrafish embryos were imaged using a Samsung 13-megapixel camera with an f/1.9 aperture applied to the ocular of Motic SMZ-171 binocular. RTPCR was used to determine gene expression in zebrafish embryos and adults. Graphical representation of the expression data and statistical analysis were conducted using the unpaired two-tailed Student's t-test with GraphPad Prism 8 software. |
