Introduction: Chronic inflammatory diseases represent constant medical, social, and economic burden, and their pharmacotherapy is still an unresolved issue. Proinflammatory stimulated macrophages generate high levels of reactive oxygen and nitrogen species that induce oxydative/nitrosative stress and tissue damage. Medical potential of chloroquine analogues is increased, among other things, due to their potent anti-inflammatory effects. Since the quinoline skeleton has great potential for the development of new drugs, a new promising approach is based on the synthesis of hybrid compounds that act on multiple targets by distinct mechanisms. Research objectives: To investigate the anti-inflammatory activity of quinoline-arylamidine hybrids in murine macrophage RAW264.7 cell line by determining the antioxidant capacity of hybrids, and the amount of nitric oxide and malonyldehyde. Materials and methods: The MTT assay was used to investigate the effect of seven quinoline-arylamidine hybrids on the growth of the murine macrophage cell line RAW 264.7 under unstimulated conditions and after proinflammatory stimulation with E. coli lipopolysaccharide (LPS). Nitric oxide (NO) and malondidaldehyde (MDA) levels were determined by spectrophotometric methods. Antioxidant capacity of tested compounds was tested by 2,2'-azino-bis(3-ethybenzthiazoline-6-sulfonic acid (ABTS) method. Results: Tested hybrid compounds differentially influenced proliferation of non-stimulated and LPS-stimulated RAW 264.7 cells. The hybrid compounds have not presented ABTS radical-scavenger activity. In the LPS-stimulated RAW 264.7 cells 10 μM compounds slightly decreased production of NO and significantly modulated malondialdehyde levels in TBARS test. Conclusion: Molecular hybrids of 7-chloroquinoline and arylamidine moieties joined through 2-aminoethanol linker may express anti-inflammatory activity by modulating the production of NO and MDA in LPS-stimulated RAW 264.7 cells.
