Liver plasma membrane lipid coposition in Sprague Dawley male rats fed with fatty and sweet food and treated with metformin or liraglutide Introduction: There is assumption that bad food consumation affects alternations in composition of cell membrane. Additionaly, animals which have diabetes also have decreased number of gangliosides what leads to assumption that decreased number of gangliosides directly affects development of diabetes type 2. Since some of antidiabetics can affect composition of cell membrane their influence was examined. Aim: Purpose of this study was to compare composition and quantity of gangliosides in liver cell membrane in Sprague Dawley male rats fed with fatty and sweet food and animals that have been given antidiabetics metformin and liraglutide along with such diet. Materials and methods: Research was carried out on tissue of 12 randomly selected animals, 3 animals from each group (males on standard diet, males fed with fatty and sweet food, males fed with fatty and sweet food who got metformin along with their diet and males fed with fatty and sweet food who got liraglutide along with their diet). Extraction of gangliosides from liver tissue was made by Svennerholm's method of organic solvents, while for their separation was used Thin-Layer Chromatography (TLC) on High Performance Thin-Layer Chromatography (HPTLC) silica-gel tiles. Results: Fatty and sweet food causes decrease in gangliosides of b and c series, so as in GM3 ganglioside, but it increases quantity of X ganglioside. Effect of metformin is reversed here, respectively metformin increases majority of gangliosides, especially GM3. Liraglutide decreases quantity of all gangliosides, while in combination with fatty and sweet food liraglutide increases only GM3. Conclusion: Fatty and sweet food affect the reduction of ganglioside expresion in cell membranes and thus change composition of cell membrane. Furthermore, antidiabetics have shown different influences on the increase or decrease in the expresion of gangliosides in cell membranes.
