This research is based on the study of the expression of the RNaseH2A gene, which plays an important role in the pathophysiological process of various types of cancer, including bladder cancer. The human RNaseH2 complex acts as a tumor suppressor because it keeps genome stability under normal physiological conditions. The aim of the study was to determine the possibility of using the expression of the RNaseH2A gene as a marker of the probability of recurrent urothelial tumor compared to a tumor that is non-recurrent. The samples were embedded in paraffin blocks from which RNA was isolated, converted to cDNA and then used for a qRT-PCR. The results obtained by qRT-PCR represent Ct (cycle threshold) values which defines the cycle in which the fluorescent signal exceeds the set detection threshold. ΔΔCt determines the relative values of changes in the target gene compared to the control and is directly proportional to the expression of the target gene in the sample. In this study, we compared three groups of tumor tissues (34 samples of tumors that would not recur (NR), 17 samples of tumors that would recur (R1), and 11 samples of tumor recurrence tissues (R2)) and measured relative changes in RNazeH2A gene expression. ΔΔCt for the previously mentioned three groups, were compared by one-way analysis of variance, ANOVA test. ΔΔCt was compared for 10 samples of R1 and R2 from the same patient. ΔΔCt values for R1 and R2 were compared by paired T-test. ΔΔCt for all three groups were not significantly different (P-value was 0.616). The ΔΔCt values for R1 and R2 from the same patients also didn’t show statistically significant difference (P-value was 0.4391). There is no difference in RNazeH2A gene expression in samples of urothelial bladder tumors that would recur compared with samples of tumor tissues that would not recur. There is also no difference in RNazeH2A gene expression in urinary bladder tumor samples that would recur and tumor recurrence tissue samples in the same patients. RNazeH2A gene expression cannot be used as a marker of the likelihood of urothelial bladder tumor recurrence compared to a tumor that would not recur.
